Cloning and Nucleotide Sequencing of the Gene (alpI) Encoding Alkaline Protease from Aspergillus oryzae

Abstract

|
Alkaline protease gene (alpI) from Aspergillus oryzae was isolated using polymerase chain reaction (PCR)-amplified DNA fragment as a probe, and a 2.6 kb ApaLI-Xbal fragment containing alpI gene was sequenced. The nucleotide sequence, except for two base pair substitution, was in accordance with other known DNA sequences. One of the substitution was within the structural gene, and the other was outside the structural gene located in 3¢¥-direction. But these substitutions had no effect on the primary sequence of alkaline protease protein (GCT and GCC code same amino acid, Ala). It was deduced that alpI gene had three short introns; 50, 59 and 56 nt in length. The alpI gene had an open reading frame of 1377 bp and a code for protein 403 amino acid residues which were comprised of signal peptide and prepro-sequence of 121 amino acids, and mature protein of 282 amino acids.
